Yes, 51 rounds of mantra jaap means 51 * 108 repeatitions and this needs to be done on a single day, however people will find it difficult to do it one go and hence you can take breaks say every 10 rounds or so and then complete the entire chanting in one night.

Diwali night is the most auspicious time for this sadhana.
